去除细胞培养基

添加染料工作溶液

培养10分钟

使用Ex/Em=490/525 nm的荧光酶标仪进行分析

重要提示：在打开瓶子之前，将所有成分加热到室温。由于该试剂与核酸结合，应将其作为潜在的诱变剂处理，并适当谨慎处理。DMSO储备液应特别小心处理，因为已知DMSO有利于有机分子进入组织。

 

溶液配制

1.工作溶液配制

Nuclear Green LCS1工作溶液配制向10 m L反应缓冲液中加入25 µL的Nuclear Green LCS1，并充分混匀。

注：10 mL 足以进行100次试验。为了获得效果，Nuclear Green LCS1染料工作液应及时使用。

注：在- 20 ° C保存任何未使用的Nuclear Green LCS1 (成分A )。

 

操作步骤

以下方案仅用于指导，具体，请按照自己的实验情况进行调整。

1.制备细胞样本并按需处理细胞。

 

2.去除细胞培养基，将细胞用PBS清洗2次。注意：去除细胞培养基是必须的，因为它可能干扰Nuclear Green LCS1的荧光。

 

3.可选：用PBS洗涤后，涉及多个时间点和样品的实验可以在- 80 ° C保存几天或进行步骤4。

 

4.加入100 µ L的Nuclear Green LCS1染料工作液，室温孵育10 ~ 60 min，避光。

 

5.使用Ex / Em = 490 / 525 nm ( Cutoff = 515 nm)的荧光酶标仪检测样品荧光。
